Personalized neoantigen cancer vaccines: An analysis of the clinical and commercial potential of ongoing development programs
Drug Discov Today. 2023 Sep 18:103773. doi: 10.1016/j.drudis.2023.103773. Online ahead of print.ABSTRACTNeoantigen cancer vaccines harbor promise as next-generation immuno-oncology therapies, whereby cancer vaccines are tailored to the patient's tumor antigen and represent the future of personalized cancer therapy. While several biotech companies have ongoing development programs, little has been published about the true commercial potential of these innovative therapies and the challenges these products will face upon regulatory approval. In this paper, we provide an overview of neoantigen cancer vaccine development progr...

Stimuli-Responsive Hydrogel Microcapsules Harnessing the COVID-19 Immune Response for Cancer Therapeutics
We present the development of stimuli-responsive microcapsules, loaded with a viral immunogenetic agent, harnessing the immune response against the Coronavirus Disease 2019, COVID-19, to selectively attack liver cancer cells (hepatoma) or recognize breast cancer or hepatoma, by expression of green fluorescence protein, GFP. The pH-responsive microcapsules, modified with DNA-tetrahedra nanostructures, increased hepatoma permeation by 50%. Incorporation of a GFP-encoding lentivirus vector inside the tumor-targeting pH-stimulated miRNA-triggered and Alpha-fetoprotein-dictated microcapsules enables the demonstration of neoplas...

Graphene oxide nanoarchitectures in cancer therapy: Drug and gene delivery, phototherapy, immunotherapy, and vaccine development
Environ Res. 2023 Aug 31:117027. doi: 10.1016/j.envres.2023.117027. Online ahead of print.ABSTRACTThe latest advancements in oncology involves the creation of multifunctional nanostructures. The integration of nanoparticles into the realm of cancer therapy has brought about a transformative shift, revolutionizing the approach to addressing existing challenges and limitations in tumor elimination. This is particularly crucial in combating the emergence of resistance, which has significantly undermined the effectiveness of treatments like chemotherapy and radiotherapy. GO stands as a carbon-derived nanoparticle that is incre...

mRNA vaccine in cancer therapy: Current advance and future outlook
Clin Transl Med. 2023 Aug;13(8):e1384. doi: 10.1002/ctm2.1384.ABSTRACTMessenger ribonucleic acid (mRNA) vaccines are a relatively new class of vaccines that have shown great promise in the immunotherapy of a wide variety of infectious diseases and cancer. In the past 2 years, SARS-CoV-2 mRNA vaccines have contributed tremendously against SARS-CoV2, which has prompted the arrival of the mRNA vaccine research boom, especially in the research of cancer vaccines. Compared with conventional cancer vaccines, mRNA vaccines have significant advantages, including efficient production of protective immune responses, relatively low s...

This study demonstrates just how vital the thymus is to maintaining adult health." « Back to Top Does Amyloid-β Aggregation Cause Broad Disruption of Proteostasis? https://www.fightaging.org/archives/2023/08/does-amyloid-%ce%b2-aggregation-cause-broad-disruption-of-proteostasis/ Researchers here speculate on the ability of insoluble amyloid-β aggregates to be broadly disruptive of the solubility of many other proteins, and thus disruptive to cell and tissue function. Is this important in aging? The evidence here shows the existence of the mechanism in a lower species, but that doesn't n...

Viral nanoparticles: Current advances in design and development
Biochimie. 2023 Aug 10:S0300-9084(23)00197-9. doi: 10.1016/j.biochi.2023.08.006. Online ahead of print.ABSTRACTViral nanoparticles (VNPs) are self-assembling, adaptable delivery systems for vaccines and other therapeutic agents used in a variety of biomedical applications. The potential of viruses to invade and infect various hosts and cells renders them suitable as potential nanocarriers, possessing distinct functional characteristics, immunogenic properties, and improved biocompatibility and biodegradability. VPNs are frequently produced through precise genetic or chemical engineering, which involves adding diverse seque...
